New colchicinoids from a native Jordanian meadow saffron, colchicum brachyphyllum: isolation of the first naturally occurring dextrorotatory colchicinoid.

چکیده
As part of our continuing investigation of Jordanian Colchicum species, the biologically active components of Colchicum brachyphyllum were pursued. Using bioactivity-directed fractionation, nine colchicinoids were isolated and characterized. One of these has a novel ring system, to which we have ascribed the trivial name (+)-demecolcinone (9), and it represents the first naturally occurring dextrorotatory colchicinoid. Another isolated compound was a new colchicinoid analogue, (-)-2,3-didemethyldemecolcine (8), while the remaining seven known colchicinoids were new to the species: (-)-colchicine (1), (-)-3-demethylcolchicine (2), (-)-cornigerine (3), beta-lumicolchicine (4), (-)-androbiphenyline (5), (-)-demecolcine (6), and (-)-3-demethyldemecolcine (7). The brine shrimp lethality test was used to direct the isolation of these colchicinoids. Moreover, all pure compounds were evaluated for cytotoxicity against a human cancer cell panel, for antimicrobial activity in an array of bacteria and fungi (including yeast), and for their potential to be allosteric modulators of the gamma-aminobutyric acid type A receptor.

As a part of our continuing investigation of Jordanian Colchicum species, the biologically active components of Colchicum hierosolymitanum Feinbr and Colchicum tunicatum Feinbr (Colchicaceae) were pursued. The brine shrimp lethality test (BSLT) was used to direct the fractionation and isolation of active components. Five and four known colchicinoids were isolated and characterized from C. tunic...
متن کاملEffect of Soluble Chitosan Application on in Vitro Germination and Growth of Dormant Corms of Colchicum
Corms and the above ground parts of the yellow meadow saffron (Colchicum luteum Baker) have been proposed for the isolation of colchicine and colchamine in our country. Corms are collected and stored in large quantities for the extraction which result in rapid depletion of the raw material sources. In order to investigate the effects of chitosan on germination and growth of corms of Colchicum l...
متن کاملFatal colchicine poisoning by accidental ingestion of meadow saffron-case report.
A 62-year-old male died of colchicine poisoning after accidental ingestion of Colchicum autumnale (meadow saffron). He ate a salad of plant with green leaves regarded as wild garlic (Allium ursinum). A few hours later he developed symptoms of gastroenteritis and was admitted to hospital. In spite of gastric lavage, activated charcoal and supportive measures, multi-organ system failure developed...
متن کاملA polymeric colchicinoid prodrug with reduced toxicity and improved efficacy for vascular disruption in cancer therapy
Colchicinoids are very potent tubulin-binding compounds, which interfere with microtubule formation, giving them strong cytotoxic properties, such as cell mitosis inhibition and induction of microcytoskeleton depolymerization. While this makes them promising vascular disrupting agents (VDAs) in cancer therapy, their dose-limiting toxicity has prevented any clinical application for this purpose....
متن کاملA fatal case of autumn crocus (Colchicum autumnale) poisoning in a heifer: confirmation by mass-spectrometric colchicine detection.
A heifer developed severe signs of acute gastrointestinal irritation 48 hr after ingesting fresh leaves of Colchicum autumnale growing on a damp meadow. Confirmation of the suspected toxicosis was obtained by detecting colchicine in serum and urine using liquid chromatography coupled with tandem mass spectrometry using atmospheric pressure chemical ionization. Although the serum colchicine conc...
